Mr. Mime faces tough competition from other Psychic-types in PU like Beheeyem, Swoobat, and Duosion but finds its niche in a superior movepool. Access to Focus Blast while other Psychics are stuck with Hidden Power means that Mr. Mime hits common switch-ins like Audino and Zweilous harder, and the ability to use Healing Wish effectively is fairly unique and valuable in PU. This deep movepool allows Mr. Mime to utilize a strong Choice Scarf set or a powerful Nasty Plot wallbreaker set. However, some walls like Bronzor can wall Mr. Mime regardless of coverage, and the sheer amount of good Psychic-types in the tier means that a Psychic-type check like Shiftry or Sneasel is a common sight. Additionally, Beheeyem's superior firepower, Duosion's better bulk, and Swoobat's faster natural Speed means that Mr. Mime can struggle to stand out against the competition.

Both Focus Blast and Signal Beam hit Dark-types like Shiftry, but Signal Beam is more reliable while hitting Psychic-types like Beheeyem and opposing Mr. Mime, and Focus Blast is stronger and hits Steel-types like Klang as well as Scraggy. Trick is the heavily preferred option for the third slot for disabling otherwise-unassailable walls such as Bronzor, but this slot can also be used for Signal Beam to have access to all coverage. Healing Wish is the solid fourth option and one of the main reasons to use Choice Scarf Mr. Mime, as a complete heal in a tier filled with status and chip damage like PU to a key teammate can easily change the direction of games. Filter is chosen as the ability to sponge some damage from the stray non-STAB Sucker Punch from a threat like Mawile that would normally OHKO with Sucker Punch but now has an unfavorable roll even with a Life Orb.
Mr. Mime even with its coverage is still stopped by certain walls such as Bronzor, so wallbreakers such as Simisear can help clear the way for a sweep later on. Mr. Mime also has to choose its coverage carefully, as getting locked into an exploitable move can be detrimental. A Mr. Mime locked into Psychic is inviting in threats like Shiftry, and getting locked into Focus Blast or Signal Beam can invite in other threats like Duosion or Muk respectively. As such, having teammates capable of responding to these switchins such as Monferno, Zweilous, and Stunfisk are key for keeping momentum on your side. Partners such as Ursaring and Monferno, who are prone to acquiring chip damage and status like Sleep or Paralysis and therefore appreciate a well-timed Healing Wish to close out games, also love having Mr. Mime on their team.

Substitute is chosen to better set up against more passive walls like Bronzor and dodge the occasional Sucker Punch from would-be threats like Shiftry. The choice between Psyshock and Psychic largely comes down to preference as most walls are 2HKOed regardless of choice, but some of the sturdiest walls in the tier like Articuno are cleanly 2HKOed by Psyshock whereas Psychic is either an unfavorable coinflip or a 3HKO. Because of this, it is recommended that if Mr. Mime chooses Psychic to also pair it with Twistedspoon and Focus Blast to be able to get through these walls anyway. That being said, the choice of attacks are largely down to what walls you want to break down (such as Duosion versus Klang as an example) and Mr. Mime will see similar results regardless. Leftovers is the preferred item for longevity and getting more uses of Substitute, but Twistedspoon can be used for bluffing the more common Choice Scarf and pushing Psychic from a roll to 2HKO on Audino to a clean 2HKO after a Nasty Plot. Soundproof is the chosen ability to block Roar from certain walls like Zweilous that would very much like to phaze Mr. Mime from behind its Substitute.
Mr. Mime is good for baiting in and breaking through walls like Audino, Zweilous, and Bronzor, so sweepers like Huntail make for good partners. Pivots like Zebstrika also make for good partners as they can bring in Mr. Mime safely for a free turn to Substitute while Mr. Mime clears out the walls that might stop the pivots. Mr. Mime also tends to attract Dark- and Psychic-types switchins like Sneasel and Duosion, so having a check to those threats like Monferno and Zweilous can be handy. While not contributing much to defensive synergy, Mr. Mime can still threaten out Fighting-types like Monferno and Machoke easily so partners like Shiftry and Stoutland can thrive.
